Can I connect a uv to 1 of my 3 drain lines?
or does it have to connect to my return.

Yes.
One of the problems with running a UV filter on a reef tank is that (if properly installed) it will kill both good and bad things.

In addition, UV filters aren't an effective treatment for ich. :-\

I would just use a union. Unless there is some need to turn off a drain. Then you would need a valve also.
